Natural de Nova Iorque, frequentou o William and Mary College e a Faculdade de Direito da Universidade de Chicago. Foi procurador-geral do Distrito Sul de Nova Iorque e procurador-geral federal adjunto durante a administração do presidente George W. Bush. No início de 2013, Comey tornou-se professor de direito e investigador em Direito de Segurança Nacional na Columbia Law School. Vive na Virgínia.
